چکیده

T he March 25 approval of ipilimumab (Yervoy) by the U.S. Food and Drug Administration was a milestone for the embattled field of cancer immuno-therapy. As the first agent to increase overall survival in a phase III melanoma trial, ipilimumab appeared to vindicate the immune surveillance hypothesis of cancer first proposed in 1957. That hypothesis, which posits that the immune system plays a key role in keeping tumors in check, gave rise to hopes that boosting the immune system could eradicate established tumors. After three decades of almost continual futility in the clinic, ipilimumab is the first clear success: It produces durable, complete responses in a small but consistent proportion of melanoma patients. But ipilimumab ' s objective response rate is only 10% – 15%, and the drug sometimes causes severe immune-related side

Microtubule dynamics and signal transduction at the immunological synapse: new partners and new connections.

Antigen recognition induces T cells to polarize towards antigen presenting cells (APC) generating an organized cell interface named the immunological synapse. T-cell microtubules (MTs) reorient the MT-organizing centre (MTOC) to the immunological synapse central region, while MT irradiate towards the synapse periphery. Martı́n-Cófreces et al (2012) describe in this issue that the MT plus-end-bin...

Numbers matter: quantitative and dynamic analysis of the formation of an immunological synapse using imaging flow cytometry.

Activation of T lymphocytes by antigen-presenting cells (APC) results in the formation of an immunological synapse. Following contact with the target cell, key signaling and adhesion molecules polarize within minutes to hours to the T cell-APC interface. Multispectral imaging flow cytometry, a new technology which combines flow cytometry with imaging, was used to visualize and quantify the recr...

The role of the cytoskeleton at the immunological synapse

It has been over 30 years since the reorganization of both the microtubule network and a 'peculiar actin polarization' was reported at the contact area of cytotoxic T lymphocytes interacting with target cells. Since that time, hundreds of studies have been published in an effort to elucidate the structure and function of the microtubule network and the actin cytoskeleton in T-cell activation, m...

Coordination of T cell activation and migration through formation of the immunological synapse.

T cell activation is based on interactions of T cell antigen receptors with MHC-peptide complexes in a specialized cell-cell junction between the T cell and antigen-presenting cell-the immunological synapse. The immunological synapse coordinates naïve T cell activation and migration by stopping T cell migration with antigen-presenting cells bearing appropriate major histocompatibility complex (...

CTLA4 as Immunological Checkpoint in the Development of Multiple Sclerosis

We investigated a patient who developed multiple sclerosis (MS) during treatment with the CTLA4-blocking antibody ipilimumab for metastatic melanoma. Initially he showed subclinical magnetic resonance imaging (MRI) changes (radiologically isolated syndrome). Two courses of ipilimumab were each followed by a clinical episode of MS, 1 of which was accompanied by a massive increase of MRI activity...
